Peptide Synthesis

Peptides are natural or synthetic compound comprising of two or more amino acids linked through  an amide formation. This covalent chemical bond formed between a carboxyl group of one amino acid to a nitrogen atom of another amino acid.  Peptides are differentiate from protein base on the number of amino acid residues. The shortest peptides are called dipeptides which consisting of 2 amino acids joint together by a single peptide bond.  Polypeptide consist of 10-50 amino acids residues are called peptides, peptides contain more than 50 amino acids often refer as protein. Peptides often classified by function or by synthesis. Peptides classified by function including but not limited to hormones, neuropeptides, HLA peptides, infectious disease, cancer peptides.

Synthetic peptides have become significant commercial and pharmaceutical products, ranging from the dipeptide sugar substitute aspartame to clinically used hormones such as oxytocin, adrenocorticotropic hormone, and calcitonin.  The development of different application using synthetic peptides including the development of epitope-specific antibodies against pathogenic proteins, the study of protein functions and the identification and characterization of proteins. Furthermore, synthetic peptides are used to study enzyme-substrate interactions within important enzyme classes such as kinases and proteases, which play a crucial role in cell signaling.

In cell biology, receptor binding or the substrate recognition specificity of newly discovered enzymes can often be studied using sets of homologous synthetic peptides. Synthetic peptides can resemble naturally occurring peptides and act as drugs against cancer and other major diseases. Finally, synthetic peptides are used as standards and reagents in mass spectrometry (MS)-based applications. Synthetic peptides play a central role in MS-based discovery, characterization and quantitation of proteins, especially those that serve as early biomarkers for diseases.
